The Centre for Research in Advanced Therapeutic Engineering wishes to purchase a Chromatography System for Fast Protein Liquid Chromatography, for analytic and preparative protein and virus purification purposes. It is essential that the Fast Performance Liquid Chromatography operating software is compatible with an established AKTA system in the Research Centre (running Unicorn) to allow for the exchange of protocols over a network system Proposed System ( Suppliers to state model type here) No Feature Indicate Complicance ( YES) or Non Compliance ( NC) Details 1.0 Technical Specifciation 1.1 Process Target Specifciation: • Essential that the FPLC operating software is compatible with established AKTA system in the Research Centre (running Unicorn) to allow for the exchange of protocols over a network system • System pumps with 25ml/min flow rate. • UV monitor to measure absorbance at 3 simultaneous wavelengths from 190- 700nm • Monitors/Flow Cells to measure pH (0 – 14) and conductivity (0.01 – 999.99 mS/cm). • Fraction collector to allow collection in deep well plates (various volumes), tubes and bottles • Sample pumps capable of >4ml sample injection • Pressure sensor for column safeguarding • Inlet valve kit capable of switching between multiple buffers and cleaning solutions • Columns allowing: o separation of proteins with molecular weight from ranges 10,000 to 600000 Da with a flow rate of > 1.5 ml/min o separation of proteins with a molecular weight of 3,000 to 70,000 Da with a flow rate of 1 ml/min o the loading of >4ml of protein sample for subsequent protein separation with a flow rate of > 1.2 ml/min o purification of histidine tagged proteins by immobilised metal ion affinity chromatography o Purification of bacteriophages to a titre of 10,000,000,000 (10e10) particles per ml • Accessories Kit including all connectors necessary for equipment use •
